Can I define some extra nodes(not connecting any beams of columns) at floor level in order to find the Floor acceleration at that node point ?
Actually I need to find the Floor acceleration at Center of Rigidity. But the point where Centre of Rigidity is not lying at any node. Also, If I define any node at floor level, what should be the SP constraint there for that node ?

It depends on the circumstances.
Assuming that you are using rigid diaphragms for your floors, you could add extra nodes in the floor, and include them in the interaction for the rigid diaphragm as slaves (of course, in that case, you need to fix the other 3 DOFs not included in the diaphragm constraint).
